The cause of Aicardi Syndrome is unknown at this time. In some cases, experts believe it may be a result of a gene defect on the X chromosome.

The location of the gene mutation responsible for Aicardi syndrome has been localized to Xp22.3. At or near this same locus is the gene responsible for micropthalmia with linear skin defects(MLS) and the gene responsible for Goltz syndrome.
